dc.description.abstract Understanding the biophysics of fluid dynamics within the context of transcriptional regulation, mediated by nuclear factor (NF)-κB, is crucial to developing a consensus on the molecular basis of fluid mechanics and imbalance. Amiloride, an antikaliuretic-diuretic agent, has recently entered the realm of NF-κB as a key player in regulating the molecular association of fluid dynamics with inflammation and oxidative stress. With the identification of flanking regions encoding the amiloride-sensitive channels that are NF-κB-responsive, a new theme emerges which underlies the significance of this association. What is the role of NF-κB in regulating fluid mechanics-is it a physiologic or immunologic function? Conversely, amiloride is purported as a major regulator of this transcriptional pathway. It is the mainstream of this survey, therefore, to outline current advances on the biophysics and nature of the interaction existing between amiloride, amiloride-sensitive channels, and NF-κB, while searching for potential molecular mechanisms. © 2004 Published by Elsevier Inc.
